"With these live disks, we can do all the things the lab teacher doesn't want us to do!" "Or we could just unplug from the network on bootup so that the "seeing eyes" program doesn't boot up so she can't see what we're doing unless she's physically here"

Theoretically, however, yes, it should work. You must be careful about the boot, however, since Grub wont be able to map to the same drive as it would with a standard system (since it's a USB device rather than a fixed HDD it wont always be hd1 as far as Grub is concerned).
